IL-6 is a pleiotropic cytokine of 20-22 kDa, crucial in inflammation, immunity, and hematopoiesis. It plays a critical role in the acute phase response by stimulating hepatocytes to produce C-reactive protein (CRP) and other acute-phase proteins. IL-6 is also involved in the differentiation and activation of T cells and B cells and thereby promoting adaptive immunity. As a crucial driver of B-cell differentiation, IL-6 induces plasma cell formation and thereby antibody production. It serves as a potent stimulator for plasmacytoma and multiple myeloma cells, contributing to their proliferation and survival. Chronic IL-6 overexpression is linked to plasma cell neoplasia, where it drives malignant transformation and sustains tumor growth.
Besides oncology, IL-6 drives autoimmune diseases such as rheumatoid arthritis and systemic lupus erythematosus (SLE). It promotes synovial inflammation and osteoclast differentiation, resulting in joint destruction in arthritis. Elevated IL-6 levels are also observed in cytokine storm syndromes, such as COVID-19-associated hyperinflammation, where an overactive immune response leads to significant tissue damage.
IL-6 is used as a culture supplement to enhance hybridoma cell growth and antibody production. Its ability to sustain long-term B-cell proliferation makes it valuable in monoclonal antibody development for research and therapeutic applications.
