Anti-FHR-3 Antibody: Clone 3.11

Anti-FHR-3: clone 3.11, is a mouse monoclonal antibody that specifically recognizes human full-length Complement Factor H-related protein 3 (FHR-3). In plasma, FHR-3 exists in various glycosylated forms, ranging from 35 to 56 kDa. FHR-3 is synthesized and secreted primarily by the liver. However, it is also secreted and synthesized by immune cells such as monocytes, macrophages, and dendritic cells. Structurally, it shares homology with Complement Factor H (CFH), a key regulatory protein in the complement system.

FHR-3 operates as part of the complement regulatory network predominantly within the alternative pathway (AP). It contributes to the control of the complement cascade, although its precise role remains unclear. Studies report weak cofactor activity, enhancement of FH cofactor activity, and minor inhibitory effects in hemolytic assays with FH-depleted serum. However, these results need to be confirmed in independent studies.

Disruptions in FHR-3 expression or function have been linked to specific diseases. For instance, atypical hemolytic uremic syndrome (aHUS), age-related macular degeneration (AMD), and systemic lupus erythematosus (SLE). In these disorders, aberrant complement activation and deposition on self-tissues can contribute to pathological processes. FHR-3’s involvement in the alternative pathway makes it a promising therapeutic target for diseases caused by complement dysregulation.
